I'm using a datfile, a new version of the dat file was released and I want to replace the datfile I am using with the new version. How do i do this without losing my settings?

Add the new datfile in the profiler. Either by drag&drop or by clicking on "Add datfile". After that it will be listed under "new / unused datfiles". As soon as you load it (doubleclick or load button), a prompt will appear.
If it's an update to a previous datfile, clrmamepro will detect that and asks you if you want to update.
If not, it either is a new datfile for a new system/emulator/whatever, or the datfile author made a mistake and the datfile-update-detection failed. If the update detection doesn't pop up, you will be still able to update a previous profile with the new dat or create a new profile.

to sum it: add the profile, load the profile...update procedure is done automatically.
