In the profiler you either add datfiles (e.g. with drag'n drop or add button), or you add an exe-based datfile with the 'Create' button. There you have to add a description and an exe name of an emulator binary e.g. MAME. Best is to use the browse button there to select your emulator binary. If an error appears, something's wrong with that exe file. Either it's locked by a different program (e.g. a virusscanner locks the access) or it's simply not working correctly. Since you're on a 64bit OS, you may want to check if the chosen executable actually works. So go and check that emulator exe and also check if it supports the known database export functions. For MAME, this is -listxml.
