I have the Romset divided in two directory, in one I hold the Romset of the Mame, while the other one includes only the differences with the Mame Plus ASH, I want to know if it were possible to set the first read-only so that the changes is done only in the second and the Romset of the official MAME is not modified.

Not really. The easier would be to use 2 profiles. One for official MAME and the other for a diff datfile for Mame Plus ASH (which can easily be created with mamediff).
Or the second holds a dat for MAMEplusAsh but includes the others paths (assuming MAMEplusAsh only holds additional sets).


anyway...imo there is not really a need to keep MAME clones... ;)
