Plus them into an Ipod or the like first to test them. Plug headphones into the main board (green plug). That'll give you a definitive answer.

Possibly a break in the wire is what I would guess if still no results with the speakers. You can folding/bending the wire while playing music and see if you get anything to confirm it.
